MR imaging and MR spectroscopy of human during physical stress (MR spectroscopy imaging, MR diffusometry, MR relaxometry etc.)

The dissertation is concerned to in vivo phosphorus MR spectroscopy (31 P MRS) and 1 H MR imaging (MRI) of muscle in combination with physical workload. The theoretical part of the thesis describes methodology of 31 P MRS measurement and its clinical use in research of metabolic changes in diabetes, heart failure and peripheral artery disease (PAD). The results of the thesis are divided into methodical and clinical parts. Methodical results deal with the construction of experimental equipment, software modification and development, and show of the reproducibility of the dynamic 31 P MRS. The MRI after exercise was used to the describe involvement of the individual calf muscles to muscle contraction during pedal movement in MR compatible ergometer. The first part of the clinical results of the thesis describes changes in muscle metabolism during diabetes and critical ischemia. In patients with critical ischemia the effect of treatment by angioplasty or transplantation of mesenchymal stem cells was evaluated. In the second part of the clinical results the metabolism of patients with heart failure complicated by sideropenia was studied. In these patients the effect of experimental treatment by iron carboxymaltose was described.

Fluorine nuclear relaxation times in chelates with lanthanides

Contrast agents containing fluorine have great prospects for magnetic resonance imaging in medicine. Low representation of fluorine in human body and the suitable magnetic properties of its nucleus 19 F, provide great sensitivity. The slow relaxation of 19 F, can be shortened by using paramagnetic complexes.In this thesis, transversal and longitudinal 19 F relaxation times of chelates with different paramagnetic lanthanides (Ce, Yb,Tm, Dy, Ho) were measured in two different magnetic fields: 4,7 and 11,7 T. Their values meet the expected assumptions. Furthermore, the values are compared with Bloch-Redfield- Wangsness theory.While Ho, Tm, and Dy reduce fluorine nuclear relaxation times to milliseconds, which is too short for MRI, relaxation rates of Ce a Yb reach approximately 100ms. Such relaxation rates can be considered as optimal for imaging methods. On the basis of our results it is possible to find suitable candidates for fluorine contrast media and optimize the measurement sequence settings in further research or in clinical use.
SMV-2015-37: Development of an animal model of septic encephalopathy for MR studies, execution and evaluation of a pilot study

Two animal models of septic encephalopathy in mice were tested: the induction by lipopolysaccharide, and the fecal model, whose objective was to test their applicability in the study of pathophysiology of septic encephalopathy by magnetic resonance in 9.4 Tesla field. Three MR imaging protocols were optimized: T2-weighted RARE for morphological imaging, protocols for native perfusion imaging ASL, and for contrast-enhanced perfusion imaging DCE applying Magnevist, whose main target was the detection of potential changes in the cerebral blood flow and alteration of the blood brain barrier function. Measurements were performed in Bruker Biospec 94/30 with a set of 27 mice anesthesized by isoflurane, whose physiological functions were continuously monitored. The original and analyzed data were provided to the client.

This paper shows the way for magnetic susceptibility determination for weakly magnetic materials giving no MR signal by means of induced inhomogeneity. The results obtained by Finite Elemnt Method (FEM) modeling as well as data measured by MRI are introduced. Properties of described method are compared with measurement based on inductive method.
